茶树鲜叶抗坏血酸过氧化物酶活性的变化规律及测定方法

摘    要:探讨茶树鲜叶抗坏血酸过氧化物酶(APX)活性的测定条件,分析不同茶树品种以及茶树鲜叶不同叶位APX活性的变化.结果表明:聚乙烯聚吡咯烷酮(PVPP)加入量约为鲜叶重的1.5倍,酶提取液pH为7.8.反应液pH为7.O,抗坏血酸(AsA)浓度为0.50 mmol/L时茶树鲜叶APX活性最高;茶树鲜叶APX活性随冷藏时间的延长呈下降趋势;福建7个主要茶树品种以福鼎大白茶的APX活性最高,铁观音的APX活性最低;茶树鲜叶不同叶位APX活性变化趋势为:芽>第一叶>第二叶>第三叶>老叶>嫩茎.

Abstract:The ascorbate peroxidase activities in the fresh leaves of tea plants were determined and their optimum conditions for determination discussed. The changes of the ascorbate peroxidase activities in the different tea cultivars and on the different leaf positions of the fresh leaf were observed. The ascorbate peroxidase activities in the tea fresh leaves were the highest when determined under the conditions that the weight of PVPP added was 1.5 times of the weight of fresh leaf, that the pH was 7.8 for extraction solution and 7.0 for reaction solution and, that the AsA concentration was 0.50mmol/L. The ascorbate peroxidase activities in tea fresh leaves showed a tendency to decrease along with cold storage duration. The tea Fuding Dabaicha was the highest in ascorbate peroxidase activities among 7 major tea cultivars in Fujian Province, while Tieguanyin was the lowest. The ascorbate peroxidase activities in the tea changed with leaf positions in the order of the bud > the first leaf> the second leaf> the third leaf> the old leaf >the young stem.
