辣椒Rop cDNA分离及其序列分析

摘    要:从cDNA文库中分离获得1个cDNA阳性克隆,测序和序列分析结果表明,其长度为1141 bp,含有1个长度为197 aa的开放读码框架,与烟草等植物的Rop有较高的同源性,该cDNA是从UV处理的辣椒cDNA文库中分离获得的,可能与辣椒适应UV辐射胁迫有关.该cDNA的分离为进一步研究其在辣椒适应UV辐射防御反应的分子机理研究奠定了重要的基础.

Abstract:Rop is a member of Rho superfamily that exists only in plants, with the transformation from GDP-bound inactive form to GTP-bound active form, and the Rop plays important molecular switch roles in adaptation of plants to environmental stresses, defense reaction as well as other life processes. In this study, a Rop cDNA positive clone of Capsicum annuum was isolated from a cDNA library constructed from Capsicum annuum leaves treated with UV by a method of PCR-based 96-hole screening. Sequence analysis showed the positive clone turned out to be 1 141 bp in length, harboring an open reading frame (ORF) of 197 aa in length. The positive clone shared a high similarity in induced amino acid sequence with that of Rop of other plants such as tobacco. The positive cDNA clone of the Rop might have specific function in the adaptation of Capsicum annum to UV radiation. Therefore, isolation of Rop positive cDNA clone lays an important foundation for further molecular mechanism study on response of Capsicum annuum to UV radiation stress.
