麦冬须根正丁醇萃取物化学成分的分离与鉴定

摘    要:为了研究麦冬须根中的化学成分,本研究采用大孔吸附树脂、ODS和Sephadex LH-20多种柱色谱技术对麦冬须根乙醇提取物的正丁醇萃取物进行分离纯化;通过波谱数据与理化性质分析并结合文献进行化合物结构鉴定。从麦冬须根的正丁醇萃取物中分离得到10个化合物,其结构分别鉴定为5-甲氧基-6-甲基-7-羟基-8-醛基-3S-(3', 4' -亚甲二氧基苯)-4-二氢色原酮(1)、甲基麦冬黄烷酮A(2)、甲基麦冬黄烷酮B(3)、大黄素(4)、肥牛木素(5)、E-对羟基桂皮酸(6)、Z-对羟基桂皮酸(7)、4-羟基苯甲酸(8)、25R-spirost-5-ene-1β, 3β-diol 1-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside](9)和β-谷甾醇(10)。其中化合物1为新的高异黄酮类化合物,命名为麦冬黄烷酮D',化合物5和7为首次从百合科植物中分离得到,化合物9为首次从该植物中分离得到。

Abstract:To study the chemical constituents from the fibrous roots of Ophiopogon japonicus, ten compounds were isolated from the n-butanol extract of the fibrous roots by a combination of various chromatographic techniques, such as macroreticular resin, ODS and Sephadex LH-20 column chromatographies. The chemical structures of the compounds were elucidated as 5-methoxy-6-methyl-7-hydroxy-8-aldehydo-3S-(3', 4'-methylenedioxybenzyl)chroman-4-one (1), Methylophiopogonanone A (2), Methylophiopogonanone B (3), Emodin (4), Celignan (5), E-p-hydroxycinnamic acid (6), Z-p-hydroxycinnamic acid (7), 4-hydroxybenzoic (8), 25R-spirost-5-ene-1β, 3β-diol 1-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside] (9), and β-Sitosterol (10) based on spectroscopic analysis, physiochemical properties and literature data. Compound 1, named as asophiopogonanone D', was isolated and identified as a new compound. Compounds 5 and 7 were isolated from the plants of the family Liliaceae for the first time, while compound 9 was obtained from this plant for the first time.
