青葙愈伤组织诱导研究

目的 筛选诱导青葙愈伤组织的最佳外植体和最佳诱导条件.方法 选取青葙饱满成熟种子培养无菌苗,以其叶片和茎段为外植体,分别接种于添加NAA、6-BA、2,4-D组合的MS培养基中诱导愈伤组织,通过正交试验筛选出诱导愈伤组织的最佳外植体和培养基.结果诱导愈伤组织的最佳外植体为试管苗茎段,MS+0.5mg/L 6-BA+0.5mg/L NAA是愈伤组织诱导的最佳培养基,其愈伤组织诱导率为92.5％.结论该研究建立的青葙的愈伤组织培养方法,为青葙的快速繁育体系及工厂化生产提供了技术参考.

Study on Callus Induction of Celosia argentea L.

Objective This study aimed to select the best explants and the best conditions for callus induction of Celosia argentea L.Methods The mature seeds of Celosia argentea L.were chosen for the culture of aseptic seedlings.The seedlings＇ leaf and stem sections were taken as the explants,which were inoculated in MS media with NAA,6-BA and 2,4-D for callus induction.The best explants and medium of callus induction were confirmed with the orthogonal test.Results The best explants are stem section,and the best induction medium is MS＋0.5 mg/L 6-BA＋0.5mg/L NAA,the callus induction rate is 92.5% with the media.Conclusion The callus culture method of Celosia argentea L.is established that will provide some technical reference for the plant＇s rapid propagation and factory production.
