大孔吸附树脂法纯化黄花草木樨总皂苷工艺的研究

摘    要:目的优化大孔吸附树脂法纯化黄花草木樨叶中总皂苷的工艺。方法:以黄花草木樨中总皂苷的含量为指标,考察了5种大孔吸附树脂对黄花草木樨中总皂苷的吸附及解吸效果,筛选出最适皂苷纯化的树脂,并对纯化工艺进行了优化。结果:AB-8大孔吸附树脂适合纯化黄花草木樨中的总皂苷,最佳洗脱条件为:上样浓度为1.0mg/ml,上柱吸附流速为2.0ml·min^-1,以70%乙醇洗脱,洗脱流速2.0ml·min^-1,洗脱体积为5BV。结论所选工艺方法简便,稳定,纯化效果好。

Optimization of the Purification Process for the Total Saponins of Melilotus officinalis

Abstract:Objective To optimize the purification process for total saponins from Melilotus officinalis by orthogonal test.Methods Orthogonal test was designed for investigating optimum purification technology for Melilotus officinalis with the content of total saponins as index. Results :The optimum purificatin condition was as follow: concentrated extract passed through AB-8 resin with eluting by 5.0 BV 70% ethanol, purification with 1.0 mg/ml extraction concentration saponins ,elution flow rate was 2.0ml·min^-1, adsorption flow rate was 2.0ml·min^-1.Conclusion :The optimal purification process was simple, stable, and effective.
