摘    要:通过农杆菌介导法将抗草甘膦基因EPSPS转入玉米自交系H99中,同时研究了农杆菌浓度、侵染时间及共生培养条件等因素对转化频率的影响。结果表明:菌液浓度OD600为0.6、侵染时间20 min、共生培养时间3 d为最佳转化条件。获得的转基因植株经PCR分析鉴定,其中7株表现阳性,证明外源基因已经整合到玉米基因组中。

Abstract:The EPSPS gene was transferred into maize inbred line H99 by Agrobacterium tumefaciens mediated transformation. Factors influenced transformation efficiency was studied, including bacterial concentration, infection time and co-cultivation time. Results showed that bacterium concentration at OD600= 0.6, 20 minutes infection time and co-culture 3 days were the optimal conditions. PCR analysis on transformation plants demonstrated that 7 of them were positive and the foreign gene was integrated into maize genome.
