农杆菌介导玉米幼胚遗传转化体系的研究

摘    要:以携带pCAMBIA1301(含GUS基因)质粒的EHA105菌株侵染玉米优良自交系齐319的幼胚,培养3 d后,借助GUS基因的瞬时表达率,对影响农杆菌介导玉米幼胚转化的部分因素进行优化。结果表明,农杆菌侵染液最佳浓度OD600为0.4~0.6,最适合用于转化的幼胚大小为直径1.5 mm,最佳侵染时间为20 min,GUS瞬时表达率为23.46%。

Abstract:An optimized transformation system is developed for maize elite inbred lines using Agrobacterium mediated gene transfer by studying various important factors that affect transformation efficiency. The system was performed by transient expression of GUS gene after explants were placed on cultivation medium for 3 days. The study with immature embryos of Qi319 showed that the optimized Agribacterium suspension was as following, the concentration of Agrobacterium in the inoculation medium was adjusted to OD600=0.4-0.6. The size of the embryos was about 1.5 mm in length. The time of transformation was about 20 min. GUS transient expression efficiency was 23.46%.
