玉米航天诱变SP3株系的遗传变异分析

摘    要:从132对SSR标记筛选出32对具有稳定多态性的引物,对3个玉米自交系K305、K169和698-3的118个航天诱变SP3株系进行遗传变异分析。结果表明,在3个自交系的诱变后代中分别检测到129、154、181个等位基因变异,每对引物分别检测出1~13、1~12、2~9个等位基因,平均每对引物分别检测到3.96、4.63、5.00个;每个位点的多态信息量(PIC)平均为0.501、0.587、0.708,变幅分别为0.061~0.930、0.129~0.935、0.092~0.914;与各自对照的遗传相似系数平均值分别为0.689 4、0.792 4、0.741 7,变幅分别为0.604 7~0.782 9、0.649 4~0.889 6、0.624 3~0.854 6。UPGMA聚类分析以0.75为尺度,将3个自交系的诱变株系分成4、5、6类。航天搭载可创造丰富的遗传变异,拓宽原品种的遗传基础,对不同玉米自交系的诱变效果存在差异,K305诱变效果最为明显,698-3次之,这些遗传变异与农艺性状有关。

Abstract:Thirty-two SSR primers that gave stable profiles in the sample of 3 maize inbred lines(K305,K169 and 698-3) and their 118 mutants were selected from 132 primers,and produced 129,154 and 181 polymorphic amplified fragments respectively.The average numbers of allele per SSR locus were 3.96,4.63 and 5.00 with a ranger from 1 to 13.The ranges of mutants’ polymorphism information content for the SSR loci of 3 maize inbred lines were 0.061-0.930,0.129-0.935 and 0.092-0.914 with averages of 0.501,0.587 and 0.708,respectively.The genetic similarities ranged from 0.604 7 to 0.782 9 with an average of 0.689 4 in K305 SP3 mutants,0.649 4 to 0.889 6 with an average of 0.792 4 in K169 SP3 mutants,and 0.624 3 to 0.854 6 with an average of 0.741 7 in 698-3 SP3 mutants compared with CK.The mutants of 3 maize lines were divided into 4,5,6 groups respectively with the scale of 0.75 by UPGMA.The study clarified that distinct variation existed among mutants,and the Spaceflight of different maize inbred-lines resulted in the different variation of which K305 is the best,and 698-3 is better than K169.
