树舌液体发酵的培养基优化

摘    要:以树舌发酵液提高离体小鼠肝组织的糖耗量为指标,通过单因素试验研究不同碳源、氮源、生长因子、pH以及无机盐对树舌发酵液生长的影响,筛选出适合的碳源、氮源及无机盐,并进一步通过正交试验确定各组分之间的相互作用。试验结果表明,树舌发酵液生长的最适培养基组成为葡萄糖5 g.L-1、甘露醇5 g.L-1、蛋白胨1 g.L-1、硫酸铵1g.L-1、硫酸亚铁0.5 g.L-1、氯化锌0.5 g.L-1、氯化钾0.5 g.L-1。树舌发酵液在28℃、150 r.min-1发酵培养5 d,发酵液糖耗量可高达到88.62%,明显高于未优化的培养基。

Abstract:The fermentation condition of Ganoderma applanatum were studied with glucose consumption of isolated liver as an index.In this paper,fermentation of glucose consumption selected by single factor for carbon sources,nitrogen sources,growth factor,inorganic salts and pH.Their interactions were decided by orthogonal test.Its optimized culture medium ingredients were glucose 5 g·L-1,mannitol 5 g·L-1,peptone 1 g·L-1,ammonium sulfate 1 g·L-1,ZnCl2 5 g·L-1,FeSO4 0.5 g·L-1,KCl 0.5 g·L-1.Glucose consumption was achieved 88.62% under the condition of 150 r·min-1 for 5 d at 28℃ through the fermentation of the optimized culture,which was significantly higher than basal medium.
