几个抗疫病性不同的辣椒材料抗病基因同源序列的分离与比较

摘    要: 根据已克隆抗病基因的保守序列设计简并引物, 对9 个不同抗、感疫病的辣椒种质材料基因组DNA 进行抗病基因同源性序列的特异PCR 扩增, 经序列测定和同源性分析发现14 个抗病基因同源性序列(RGAs) 与辣椒抗疫病作用相关, 其中B 引物扩增的8 个RGAs 与烟草抗花叶病基因N 、拟南芥抗丁香假单菌基因RPS2 和亚麻抗锈病基因L6 的同源性较高, 属于广谱(细菌、病毒、真菌) 抗病基因同源序列; C 引物扩增的6 个RGAs 与番茄抗叶霉病基因Cf2 和Cf9 有较高的同源性, 与抗疫病作用密切相关。研究还发现, 高感疫病的辣椒基因组中也存在抗疫病相关RGAs , 这与已报道的辣椒抗疫病微效QTLs 位点则来于感病亲本基因组的研究结论一致。

Abstract:Numerous fragments were cloned from 9 different resi st ant/susceptible pepper (Capsicum annuum) varieties to Phytophthora bligh t, using a PCR approach with degenerate primers designed from conserved motifs f ound in a number of plant resistance genes. After sequencing and homology analys is, 8 RGAs amplified by primer B were high identified with N, RPS2, L6 and b elonged to broad-spectrum RGA, 6 RGAs amplified by primer C were high identifie d with Cf2, Cf9. 14 RGAs amplified by both primer B and primer C were relati ve to function of resistancee to Phytophthora blight. RGAs derived from susc eptible lines were also associated with function of resistant to Phytophthora blight, which was accorded with reported viewpoint that resistant minor QTLs d erived from susceptible parents. These pepper RGAs could be regarded as candidat e sequences of disease resistance genes in Capsicum annuum using cloning res istant genes and molecular markers.
