摘    要: 利用口蹄疫病毒2A序列将湖北海棠MhTGA2转录因子和几丁质酶基因(Mhchitl)两个抗病相关基因融合在同一个开放阅读框下,构建二价融合植物双元表达载体,命名为MhT2AC。将该载体转化烟草,获得了8个转基因株系,MhTGA2基因和Mhchitl基因在烟草中共表达,并且转基因株系中NtSOD、NtAPX和NtPPO等抗逆基因的表达量均加强;转基因烟草T1代植株抗盐性增强。

Abstract:The plant binary expression vector containing the divalent genes of MhTGA2 and Mhchit1 isolated from Malus hupehensis and fused in a single open reading frame(ORF)by Foot and mouth disease virus(FMDV)were constructed.The vector was then introduced into tobacco via Agrobacterium-mediated.The expression of NtSOD,NtAPX and NtPPO genes were detected and salinity tolerance was evaluated in the tobacco transformants.The results showed that we successfully constructed a plant binary expression vector with bivalent fusion and denoted it as MhT2AC.The MhTGA2 gene and the Mhchit1 gene were co-expression in the transformants.Comparing with the controls,the expression of NtSOD,NtAPX and NtPPO genes were enhanced in transgenic lines.Moreover,the salinity tolerance of the T1 transformants were significantly improved.
