Transformation of Malus × domestica with MhRAR1 and MhSGT1 Genes from Malus hupehensis for Resistance of Botryosphaeria berengeriana

Abstract:
RAR1 and SGT1 were identified as required components in some signaling pathways mediated by disease resistance(R)genes in many plant species,and they function between the downstream of R protein and reactive oxygen. To understand the expression characteristics and resistant function of MhRAR1 and MhSGT1 in susceptible variety,the two genes were transferred respectively into the Fuji apple by Agrobacterium tumefaciens-mediated method. After PCR and RT-PCR amplifications on Hyg positive plants,6 MhRAR1 strains and 3 MhSGT1 strains were screened. Infected by Botryosphaeria berengeriana,the transgenic plants displayed more rapidly increase on all antioxidant enzymes(SOD/POD/CAT)activities and gentle changing waves of MDA content. It further illustrated that RAR1 and SGT1 act on the reaction of reactive oxygen species and the process of membrane reaction,as well as promote the resistant to external infringed reaction in apple.
