摘    要:以大叶朴成熟叶片为试材,采用4种改良CTAB法提取大叶朴基因组DNA,并比较其提取效果。结果表明:用这4种方法均可获得没有发生褐变、完整性好的DNA。其中方法2(用了低浓度乙醇和高盐去多糖)和方法4(用了低浓度乙醇去多糖)去多糖的效果更好一些,但获得的DNA浓度较低;ISSR扩增结果表明,4种方法提取的DNA均可用于ISSR扩增,但方法2和3扩增的效果稍好一些。

Abstract:Four improved CTAB methods were used to extract genomic DNA from mature leaves of Celtis koraiensis Nakai and the extraction effect of the 4 methods was compared.The results showed that the 4 methods obtained no browning and intact genomic DNA.The method 2(low concentration ethanol and high salt were used to remove polysaccharide) and 4(only low concentration ethanol was used to remove polysaccharide)had the better effect of removing polysaccharide but with lower DNA concentration than the method 1 and 3.Th...
