摘    要:以国家种质资源圃(北京)编目的558份桃品种的18个形态学和农艺学性状为基本数据,研究了桃品种初级核心种质构建的取样策略,包括总体取样比例的确定及取样方案的选择,以获得最佳的初级核心种质。本试验共设10%~90%9个总体取样比例;取样方案研究包括分组原则和组内取样比例的确定。结果表明,桃初级核心种质的适宜总体取样比例为10%;按品种类群分组结合多样性比例取样为构建桃初级核心种质的最佳取样方案;利用此取样策略从558份桃品种中提取56份作为核心样本,对其代表性进行检测表明所构建的初级核心种质能够很好地代表桃原始种质的遗传多样性。

Abstract:Total of 558 accessions of peach germplasm collected from the Beijing National Peach Germplasm Repository and 18 morphological and agronomic traits of them were used for studying the optimal sampling strategy for primary core collection of peach Prunus persica (L.) Batsch.]. In order to acquire the appropriate primary core collection, we developed a simulation study to identify a suitable entire sampling ratio and sampling scheme. 9 entire sampling ratios were tested and the sampling scheme following stratification into three levels, including the grouping principle, decision on sampling proportion within group and sampling method within group were studied. The results showed that 10% should be the appropriate entire sampling ratio for primary core collection of peach. Under 10% entire sampling ratio, the optimal sampling scheme was grouped mainly based on cultivar group combining with sampling proportion within group based on proportion of genetic diversity (G) when random sampling method applied within group. Using this sampling strategy to acquire the primary core collection of 56 accessions from 558 accessions of base collection of peach cultivars, and the primary core collection could represent the genetic diversities of the entire variety collection.
