杨梅雌、雄种质遗传关系的RAPD和ISSR分析

摘    要:为获得与主栽的杨梅品种遗传基础相近的杨梅雄株,文章采用了22个多态的RAPD引物和7个多态的ISSR引物,对浙江杨梅的栽培品种东魁、荸荠种、晚稻杨梅、丁岙梅和早色等10个雌株与17个雄株种质资源间的遗传关系进行了研究。结果表明,不同杨梅产地收集的这17个杨梅雄株种质资源中,雄株与品种间的遗传关系呈规律性,即同一产区的地方品种与当地的雄株间遗传相似程度最高,而同一地区不同雄株与当地主栽品种间的遗传相似程度也有不同。通过UPGMA法进行聚类分析,上述5个杨梅主栽品种分别获得了与其亲缘关系相对接近的雄株,成对遗传相似系数最高为0.958,最低为0.878。

Abstract:Information on the genetic relationship between male plants and female cultivars is required for cross breeding in Myrica rubra,which is a dioecious fruit tree mainly native to China.To find out the pairing male plants and female cultivars with near genetic relationship,total 22 polymorphism RAPD primers and 7 ISSR primers were adopted for analyzing the genetic relationship between 10 red bayberry cultivars including Dongkui(DK),Biqizhong(PQZ),Wandaoyangmei(WD),Dingaomei(DAM),and Zaose(ZS),5 most popular cultivars planted in Zhejiang province,and 17 male plants collected from 5 sites where aforementioned 5 cultivars are natively grown.Results showed that the genetic relationship between male plants and female cultivars was quite similar by comparing the genetic similar coefficients(GSC),namely GSCs between the male plants and the local female cultivars from a same place were higher than the other pairing male plants and female cultivars,and different male plants from the same area also showed different GSC value to their corresponding female cultivars.Male plants with near genetic relationship were obtained for each of the above mentioned 5 cultivars by UPGMA cluster analysis.2 putative DK male plants,HY3 and HY4,were found with GSC being 0.928 and 0.908,respectively;4 putative PQZ male plants,CX1,CX2,CX3 and CX5,were found with GSC being 0.908,0.932,0.921,0.958;2 putative WD male plants,DH1 and DH4,were found with GSC being 0.936,0.950,respectively;3 putative DAM male plants,DA2,DA3 and DA5,were found with GSC being 0.912,0.921,0.955;and 3 putative ZS male plants,XS1,XS2 and XS3,were found with GSC being 0.880,0.878,0.884.The highest GSC was 0.958,and the lowest GSC was 0.878.The results may offer helps for the selection of male parents during cross breeding in M.rubra.
