毛竹居群遗传多样性的RAPD分析

摘    要:利用RAPD技术,对来自不同地区的18个毛竹居群遗传多样性及居群间的关系进行了研究.从184条10碱基随机引物中筛选到14条能稳定产生多态标记的引物,共扩增出129个位点,主要集中在400~1 400bp之间,其中多态性位点有101个,比率高达78.3%,Shannon多态性指数较高平均为0.377.根据POPGENE32软件分析和聚类分析,计算各居群间的遗传相似性I和遗传距离D,遗传距离的变异范围为0.081~0.503,遗传相似性变异范围为0.605~0.923,表明各居群间的遗传距离和遗传相似性存在着较大的差异.地理上较近的居群多聚在一起,表现出比较明显的地域性,说明各居群的亲缘关系与地理分布有着密切的关系.

Abstract:RAPD markers were used to assess the genetic diversity and to estimate genetic relationships among the 18 populations in difference regions.The 14 primers were screened from184 arbitrary primers,and a total of 129 loci were amplified;the length of fragments was between 400bp and 1 400bp,among which,101were polymorphic and accounted for 78.29%.The Shannon's information index gene diversity were 0.377 3.The genetic distance and cluster analysis showed that the genetic identity and genetic distance were obviously different,and indicating the significant correlation between geographical distribution and genetic distance.
