黄金蒲桃组织培养初步研究

摘    要:以黄金蒲桃的茎段为外植体,研究不同质量百分比的氯化汞溶液、不同pH、不同季节取材和不同取材部位对外植体污染率和褐变率的影响,以及不同基本培养基,不同取材部位、不同质量浓度植物生长调节剂组合对芽诱导率和丛芽生长的影响。结果表明,氯化汞的最适质量百分比存0.1%~0.5%之间,最适pH为6.0,冬季取材较好,WPM和Anderson培养基对黄金蒲桃的芽诱导影响差异不大,茎段是比较理想的取材部位,培养的最适植物生长调节剂组合为0.5mg/L6一BA+0.1mg/LNAA,其芽诱导率高达91.3%,每个茎段平均丛生芽数为3.2。

Abstract:The stem segment was selected as explant to study the effects of different conditions on the tissue culture of Xan- thostemon chrysanthus. The results were as follows, the optimum concentrations of HgC12 solution ranged from 0.1% to 0. 5% ; the optimum pH was 6.0; the optimum season of material collection was winter; the influences of WPM and Anders- son Medium on the bud induction had no difference; stem segment was the ideal explant; the optimum plant growth regula- tor combination was 0.5 mg/L 6-BA + 0.1 mg/L NAA, in which bud induction rate reached 91.3% , and the average number of inducing buds of per stem segment got 3.2.
