摘    要:通过单因素试验,对红松SRAP—PCR反应体系的主要影响因子(Taq DNA聚合酶、Mg^2+、模板DNA、dNTPs、引物浓度)进行了优化筛选,得出红松SRAP—PCR反应的最佳体系:20μL反应体系中,模板90ng,Mg^2+1.0mmol/L,dNTPs0.15mmol/L,Taq DNA聚合酶1.5 U,引物0.3~0.4μmol/L。

Abstract:Optimum selection were conducted on the major influenfial factors (Taq DNA polymerase, Mg^2+, template DNA, dNTPs and concentration of primer) for the SRAP-PCR reaction systems of Pinus koraiensis by means of one-factor experiments, and the best SRAP-PCR reaction systems of Pinus koraiensis were worked out: in the 201aL reaction systems, there were 90 ng of template, 1.0 mmol·L^-1 of Mg^2+, 0.15 mmol·L^-1 of dNTPs, 1.5 U ofTaq DNA polymerase and 0.3 - 0.4 μmol · L^-1 of primer.
