摘 要: | 通过SDS-PAGE分析毒性和无毒性织纹螺肌肉蛋白的电泳图谱差异,综合比较蛋白条带的迁移率及浓度得出,毒性织纹螺肌浆蛋白的特征条带主要为23.6ku和35.0ku,无毒性织纹螺的特征条带主要为22.8ku和31.2ku;毒性织纹螺肌原纤维的特征条带主要为38.0ku、50.8ku和135.0ku,无毒性织纹螺的特征条带主要为36.9ku和125.8ku.结合优化样品制备、等电点聚焦程序等条件,初步建立了织纹螺肌肉蛋白的双向电泳条件,并通过双向电泳分析了毒性和无毒性织纹螺肌肉中存在的差异蛋白.

Abstract: | The differences of sarcoplasmic and myofibrillar proteins between toxic and non-toxic Nassarius were analyzed by SDS-PAGE and two-dimensional electrophoresis. Comparative analysis of protein patterns was made according to mobility and relative concentration of protein bands between the toxic and non-toxic Nassarius. Specific bands of the sarcoplasmic proteins such as 23. 6 ku and 35. 0 ku could be identified in toxic Nassarius,while specific bands of 22. 8 ku and 31. 2 ku existed only in the non-toxic ones. For myofibrillar proteins,specific bands of 38. 0 ku,50. 8 ku,and 135. 0 ku were found in toxic Nassarius,while specific bands of 36. 9 ku and 125. 8 ku were in the non-toxic ones. By optimizing a series of factors,such as sample preparation and focusing parameters,a 2-DE protocol for Nassarius sample was primarily established. Under this condition,two different spots were identified between the toxic and non-toxic samples. Further identification of these different spots such as by LC MS/MS would facilitate the establishment of more effective methods for differentiation of toxic and non-toxic Nassarius. |
