枇杷品种早钟6号与解放钟、森尾早生亲缘关系的RAPD分析

摘    要:应用43个随机引物对早钟6号、解放钟和森尾早生3个枇杷品种基因组DNA进行RAPD分析,共得到255条扩增带,其中82条为共同带,单态率达32.16%,表明3个品种亲缘关系密切.对子代与父、母本扩增带中共同带的分析表明,双亲的RAPD特征均在子代中表现.用引物S71进行RAPD分析,3个品种共检测出7条扩增带,其中780bp的条带是母本解放钟的特征带,1500bp的条带是父本森尾早生的特征带,在子代早钟6号中这两条特征带同时存在,且结果稳定,在DNA水平上证实了早钟6号是解放钟、森尾早生的有性杂交后代.

Abstract:Genomic DNA of three loquat cultivars (Zaozhong 6, Jiefangzhong and Moriowase) was extracted for RAPD analysis with 43 random primers. The results showed that 255 bands were amplified from the 3 cultivars, among which 82 bands were common with the monomorphic degree of 32.16%. This suggested that genetic relationship was close among the three loquat cultivars. Parents'characteristic RAPDs emerged in filial generations by comparison with parent's common bands. 7 amplifying bands were detected in the 3 cultivars by RAPD analysis with S_(71) primer. Among them, 780 and 1500 bp bands were male (Jiefangzhong) and female (Moriowase) parents' characteristic bands, respectively. Both the two characteristic bands occurred stably in filial generation (Zaozhong 6). The above results confirmed that Zaozhong 6 was the hybrid between Jiefangzhong and Moriowase on DNA level.
