紫外线辐射的葡萄幼果cDNA文库的构建

摘    要:以紫外线辐射的葡萄幼果为材料提取总RNA,用SMARTTMcDNA文库构建试剂盒构建cDNA文库,初始文库含有1.5×106个独立的单克隆,扩增后滴度达到1.2×108pfu.mL-1.随机挑取噬菌斑进行PCR检测,发现重组率在92.9%以上.

Abstract:The total RNA was isolated from young fruits of grape challenged with ultraviolet radiation,and with the isolated total RNA,a cDNA library with more than 1.5 ×106 individual clones was constructed with smarttm cDNA construction kit.The titer of the amplification cDNA library was more than 1.2×108 pfu·mL-1 and the recombinant ratio was more than 92.9%.
