摘    要:IRAP(Inter Retrotransposon Amplified Polymorphism)是一种基于反转录转座子的分子标记技术。本文应用几种Copia类型的反转录转座子进行引物设计,封35份茄子栽培品种进行了基于IRAP的遗传多样性分析。根据Tnt1,Tto1,Tnd-1和Bare-1设计的6个引物共扩增出195条带,多态性带179条,多态性比率平均为91.8%,每个引物检测到多态性带25.33条,平均为29.8条。Nei’s遗传相似系数在0.3540-0.7752之间,平均相似系数为0.5901。多维尺度分析从分子水平将供试品种分为4个类群。

Genetic diversity analysis of eggplant varieties using irap molecular marker based on retrotransposons

Abstract:IRAP (Inter Retrotransposon Amplified Polymorphism) is a kind of molecular markers based on retrotransposons. In this study, several Copia type retrotransposons have been used for designing primers, the genetic diversity of 35 eggplant varieties was examined by IRAP molecular marker. Results showed that 195 bands were amplified with six primers designed according to the sequences of Tnt1, Tto1, Tnd-1 and Bare-1, in which 179 bands were polymorphie, the ratio of polymorphie bands was 91.8%, the number of polymorphie bands per primer ranged from 25 to 33, averaging at 29.80. Nei's similarity coefficients among the tested varieties ranged from 0.3540 to 0.7752, averaging 0.5901. The relationships among the varieties were evaluated by Multidimensional Sealing (MDS), showing that the tested eggplant varieties can be classified into four groups on the molecular level.
