磷对平邑甜茶根系质膜H~+-ATPase活性的调控

摘    要:用含NaH2PO4浓度分别为0、0.2、0.4、0.6、0.8、1.0、1.2 mmol/L的营养液处理后,平邑甜茶根系质膜H+-ATPase的活性随着根区磷浓度的升高而升高,并在1.0 mmol/L处达到峰值;该酶的对硝基苯磷酸(PNPP)水解活性也明显升高,其Km值由3.48 mmol/L下降至0.97 mmol/L;同时结合抑制剂试验,钒酸钠对幼苗根系质膜H+-ATPase的抑制效应也得到了加强。

Abstract:After treated with the nutrient solution containing 0、0.2、0.4、0.6、0.8、1.0、1.2(mmol/L) NaH_2PO_4 respectively,the activity of root plasma membrane H~+-ATPase in Malus hupehensis(Pamp) Rehd.was obviously enhanced and reached the peak value at 1.0 mmol/L.PNPP hydrolysis activity was also promoted by phosphor,and its Km value decreased from 3.48 to 0.97 mmol/L.And it was also found that 1.0 mmol/L NaH_2PO_4 could raise the inhibition of H~+-ATPase activity by sodium vanadate.These results indicated that phosphor might regulate the activity of H~+-ATPase by means of affecting the phosphatase domain of this enzyme.
