颗粒细胞对山羊卵泡卵母细胞体外成熟和体外受精的影响

摘    要:试验系统研究了颗粒细胞对山羊卵泡卵母细胞体外成熟和体外受精的影响。COCs、机械裸卵和自然裸卵的体外成熟率分别为68.3%、45.6%和6.1%,组间差异显著(P<0.05),卵裂率分别为41.5%、8.1%和0,差异显著(P<0.05)。对照组和15×104个/ml、150×104个/ml颗粒细胞组卵母细胞体外成熟率分别为65.9%、60.2%和64.6%,无显著差异(P>0.05),但均显著高于1 500×104个/ml(50.0%)和1.5×108个/ml(30.3%)颗粒细胞组(P<0.05)。颗粒细胞单层共培养系统卵母细胞成熟率为75.8%,显著高于150×104个/ml颗粒细胞条件培养液组(63.4%)和M199培养液组(66.7%)(P<0.05)。颗粒细胞单层共培养系卵裂率为50.0%,显著高于150×104个/ml条件培养液组(36.7%)和M199培养液组(38.2%)(P<0.05)。

Abstract:The experiment was conducted to study the effects of granulosa cells on in vitro maturation(IVM) and in vitro fertilization(IVF) of goat follicular oocytes.The IVM rates of COCs、denuded oocytes and naked oocytes were 68.3%,45.6% and 6.1% respectively.There was significant difference among all groups.The cleavage rates were 41.5%,8.1% and 0 respectively.The cleavage rates of COCs was significantly higher than that of denuded oocytes and naked oocytes.The IVM rates of oocytes cultured in control group,15×10~4cell/ml and 150×10~4cell/ml granulosa cells were 65.9%,60.2% and 64.6% respectively,and were significantly higher than that of 1 500×10~4cell/ml(50.0%) and 1.5×10~8 cell/ml(30.3%) granulosa cells groups.The maturation rate of oocytes in monolayer co-cultured medium was 75.8% and was significantly higher than that of 150×10~4cell/ml and M199.The cleavage rate of oocytes in monolayer medium was 50.0% and was significantly higher than that of 150×10~4 cell/ml and M199.
