应用RT—PCR检测辣椒上的黄瓜花叶病毒

摘    要:根据黄瓜花叶病毒(CMV)的外壳蛋白基因序列设计、合成引物,运用两步法RT—PCR与一步法RT—PCR都可以从感病组织中扩增出与预期的780bp大小一致的目标片段。两种方法相比,一步法的特异性强、灵敏度高、省时经济、程序简单,达到了快速检测的目的。应用一步法RT—PCR对采集于山东辣椒产区的病毒病样本进行检测,结果表明6个采样地点的样本中均检测到CMV,在所采集的30个病毒病样本中CMV所占的比例为33.3%,说明CMV是山东省辣椒病毒病主要毒源之一。

Abstract:A pair of specific primers was designed and synthesized based on the nucleotide sequence of CMV coat protein genes reported before.The expected product(780 bp DNA fragment) was gained by one-step RT-PCR and two-step RT-PCR from the infected samples.Compared with two-step RT-PCR,one-step RT-PCR showed strong specificity,high sensitivity,simple procedure,economical and saving time,so it could meet short-term test.Thirty pepper samples with virus symptom collected from Shandong area were tested by one-step RT-...
