摘    要:以“舞姿”苹果与平邑甜茶的杂交后代个体为试材,结合工作实践及当前最新研究进展,对AFLP分析过程中模板DNA的制备、基因组DNA的酶切与连接、酶切连接产物的预扩增与选择性扩增、扩增产物的变性、聚丙烯酰胺凝胶电泳等过程中易出现的问题及其解决方法作了对比研究,建立了一套适合于标记苹果农艺性状的AFLP分子标记体系。该体系在应用于苹果的AFLP研究时,能够得到清晰的遗传标记图谱,试验结果重复性好,稳定可靠。

Abstract:The AFLP(Amplified Fragment Length Polymorphism) analysis system was established suited for marking agronomic characteristics of apple with the population of F_1 progenies of "Maypde"× "Muls".Common questions in the analyzing procedure including the preparation and purification of genomic DNA,restriction-ligation of DNA,pre-amplification and selective-amplification of the products of restriction-ligation,and electrophoresis on denaturing polyacrylamide gels,and their corresponding solutions were studied and summarized.The results indicated that clear genetic marker map could be drawn out with this AFLP analysis system,and the results were steady and credible,and could be easily repeated.
