屠宰山羊卵母细胞的体外成熟培养

屠宰白山羊卵巢在２５～３５℃下于２～３小时运回实验室。每个卵巢获可培养卵丘细胞－卵母细胞复合体（ＣＯＣ）０４２个（１９３４５６）；可培养ＣＯＣ在Ｍ１９９＋ｈＣＧ１ＩＵｍｌ＋１７－β－雌二醇１μｇｍｌ＋牛血清白蛋白１０ｍｇｍｌ＋庆大霉素５０ＩＵｍｌ的培养微滴中于５％ＣＯ２、空气、饱和湿度或标准气（５％ＣＯ２、５％Ｏ２、９０％Ｎ２）、饱和湿度，３９℃培养２４～２６小时，卵母细胞成熟率为９０３％（１８６２０６）。成熟卵母细胞体外受精率为７１５％（１３３１８６）。结果表明此培养系统适合于屠宰白山羊卵母细胞的体外成熟培养

In Vitro Maturation of Oocytes from Slaughtered Goats

Ovaria from slaughtered white goats were carried to a laboratory at 25～35℃ in 2～3 hours.From an ovary could be obtained 0.42 COCs.The COCs were cultured in droplets of M199 supplemented with hCG(l IUml),17-β-E 2(1 μgml),BSA(10 mgml) and gentamicin (50 IU ml)in saturated humidified atmosphere of 5% CO 2 in air or 5% CO 2,5%O 2 and 90%N 2,at 39℃,for 24～26 hours.As a result the oocyte maturation tate was 90.3%(186206) and in vitro fertilization rate of the matured oocytes 71.5%(133186),showing that the activity of the oocytes was no afected by 2～3 hours' transportation,and this culture system was suitable for in vitro maturation of oocyted from slaughtered white goat.
