摘    要:以薹菜子叶-子叶柄为外植体,研究了根癌农杆菌介导的遗传转化技术。用含GUS和NPTⅡ基因的农杆菌侵染子叶-子叶柄,获得了具有卡那霉素抗性的再生植株。再生植株经GUS组织染色,证实外源基因转化到植物基因组中。卡那霉素敏感性测定结果表明,当培养基中含卡那霉素(Km)15 mg/L时,非转化再生植株全部白化。转基因抗性芽筛选的适宜的卡那霉素浓度为10 mg/L。

Abstract:A genetic transformation system mediated by Agrobacterium tumefaciens was developed for Brassica campetris L.ssp.chinensis Makino var.tai-tsai Hort.using petioles with cotyledon as explant.The Agrobacterium tumefaciens carrying GUS and NPTⅡ genes was used for transformation.The plantlets resistant to kanamycin were obtained.GUS histochemical assay proved that GUS gene had been stably integrated into the chromosomal genome of Brassica campetris L.ssp.chinensis Makino var.tai-tsai Hort.The test showed that all the nontransformed shoots turned white on the medium with 15 mg/L kanamycin.Therefore,10 mg/L kanamycin was suitable for the selection of transformed shoots.
