摘    要: 以褐飞虱(Nilaparvata lugens Stal)为材料,分别采用SDS法,CTAB法,冰KAc法和饱和NaCl法提取褐飞虱的基因组DNA。通过电泳、紫外光谱分析和ISSR-PCR扩增等检测手段,比较分析了DNA数量与质量。结果表明: CTAB法和SDS法提取的DNA质量明显优于冰KAc法和饱和NaCl法,适于PCR扩增。并且SDS法提取的DNA经过PCR扩增后得到较多的多态性位点。因此,SDS法是实验室提取稻飞虱基因组有效,经济实用的方法。

Abstract:Genomics DNA were extracted from the rice planthopper Nilaparvata lugens (Stal) using SDS, CTAB, KAc and NaCl method, respectively. DNA was detected by electrophresis, UV and ISSR PCR. The results suggested that, the quality of DNA samples extracted by CTAB method and SDS method was obviously better and more suitable for PCR than that by the other two methods, furthermore, SDS method presented the most polymorphic sites. Therefore, the SDS method was the most effective, practical and economic method in the genomic DNA extraction of planthopper in laboratory.
