鸭出雏前后腿肌IGF-1基因表达规律研究

摘    要: 为探讨家鸭出雏前后腿肌类胰岛素样生长因子(IGF-1)的表达规律,本研究采用荧光定量PCR和原位杂交(ISH)技术,选取高邮鸭和金定鸭13,17,21,25,27日胚龄和7日龄共192个腿肌样本为研究对象。首次发现IGF-1 mRNA在两个品种不同胚龄和日龄腿肌中的表达量规律:两个品种鸭腿肌IGF-1 mRNA表达量差异不显著(P>0.05);腿肌13日胚龄表达量极显著高于其他时间点(P<0.01),并随着胚龄的增加逐渐降低。同时,原位杂交结果显示,血管周围及腿肌肌肉肌束间的肌膜中IGF-1 mRNA的表达量明显高于周边腿肌肌肉组织。研究结果表明,IGF-1基因对鸭胚胎期腿肌的生长发育具有重要意义。

Abstract:In this study, Gaoyou duck and Jinding duck were used as the test materials and we studied the expression of insulin-like growth factor (IGF-1) gene in the leg muscle from different embryonic stage using fluorescence quantitative PCR and in situ hybridization. The sample of leg muscle were from two duck breeds at the age of embryo day (ED) 13, 17, 21, 25, 27 and 7 day. The results showed that there was no significant difference about the gene expression between two ducks (P>0.05), the expression of IGF-1 mRNA at ED13 was significantly higher than the others and the expression decreased with increased age(P<0.01). The in situ hybridization (ISH) results showed, IGF-1 mRNA was localized to perisinusoidal cells and vascular cells of leg muscle. IGF-1 in the leg muscle of duck embryo maybe play an important role during embryogenesis.
