绿色木霉 X-13株固态发酵产酶优化条件的研究

摘    要: 对绿色木霉X-13株固态发酵产纤维素酶、半纤维素酶及果胶酶的优化条件进行了研究;通过综合评分的决策方法分析表明,X-13株的产酶优化条件是:稻草粉与麸皮的比例1.5∶1,料水比1∶1.5,氮源是2.0%~2.5%硫酸铵,培养基起始pH值6.0~6.5,250 mL三角瓶中装培养基15~20 g,培养时间60 h;以0.01 mol/L,pH 7.4 PBS于室温下浸提发酵培养基2 h,所得粗酶液活性最高。

Abstract:The studies were conducted to explore the optimum conditions of X-13 solid state fermentation to produce cellulase,hemicellulase and pectinase. The analysis of the decision method based on the synthetic grade indicated that the optimum conditions were listed as follow: the proportion of straw powder to wheat bran was 1.5∶1,the proportion of materials to water in the medium was 1∶1.5,the nitrogen source was 2.0%~2.5% (NH4)2SO4 and 15~20 g media were filled in 250 mL flask with medium pH 6.0~6.5, 60 h culture time and (30±1) ℃ culture temperature. The enzyme solution with the highest activity was extracted and fermented with 0.01 mol/L,pH 7.4 PBS at the room temperature for 2 h.
