水稻地方品种月亮谷与LTH叶片蛋白质的双向电泳分析

摘    要: 为了研究水稻地方抗病品种月亮谷与感病品种丽江新团黑谷(LTH)在未接种稻瘟菌前蛋白质间的差异,利用双向电泳技术对月亮谷与LTH叶片的蛋白质组分进行研究。试验选用pH 4~7,17cm的IPG胶条,利用双向电泳技术分离上述两个水稻品种的稻苗叶片的蛋白质,分析其蛋白质点表达量的差异,同时选取差异显著的蛋白质点进行质谱分析。利用Gene ontology软件对这些蛋白质进行了功能分类。月亮谷与LTH叶片蛋白的双向电泳图谱得到34个差异蛋白点,对其中20个蛋白点进行质谱测定,鉴定到13个差异蛋白,包括1,5-二磷酸核酮糖羧化酶/加氧酶大亚基,3-磷酸甘油醛脱氢酶,ATP合酶,UDP-葡萄糖焦磷酸化酶,铁氧蛋白-NADP-还原酶,叶绿素a/b结合蛋白等。因此认为月亮谷与LTH叶片中主要是参与生长代谢和防御相关的蛋白质存在差异。这些蛋白在水稻与稻瘟病菌互作中的表达模式和对抗病性的贡献值得下一步深入研究。

Abstract:In order to study the differences between proteins in rice landrace Acuce and Lijiangxintuanheigu (LTH),two dimensional electrophoresis (2 D) was used to study the protein components. Leaf proteins of these two varieties were separated through pH 4~7, 17cm IPG strips. After 2 D, the significant expression difference of spots was analyzed and identified by MS. The functional classification of these proteins was analyzed by Gene Ontology program. Thirty four differential expression proteins were detected on 2 D map compared leaf protein between Acuce and LTH. Among them, 20 spots were subject to MS analysis and 13 unique protein sequences were retrieved. The sequenced protein included ibulose 1, 5 bisphosphate carboxylase/oxygenase large subunit, glyceraldehyde 3 phosphate dehydrogenase, ATP synthetase, UDP glucose pyrophosphorylase, ferredoxin NADP reductase, and chlorophyll a/b binding protein. Therefore, the difference of proteins between Acuce and LTH mainly involved in defense and metabolism. The expression pattern and function of these protein coding genes contributing to resistance in rice rice blast fungi  interaction are valuable further study.
