云南一株高产纤维素酶菌株YN-2的筛选及其产酶条件的研究*

摘    要: 从云南农田土壤材料中分离得到一株高产纤维素酶真菌YN-2,对其进行形态及生理生化特征初步鉴定及通过ITS基因片断序列分析后,初步确定该菌株为草酸青霉。产酶条件及酶活力特性分析发现该菌株在培养4d后,羧甲基纤维素酶(CMCase)酶活达61.50U/mL, 滤纸酶 (FPAse) 酶活达19.37U/mL;酶促反应的最适反应温度为 50℃;pH值为4.8时,CMCase 达52.60U/mL, FPAse 活力达18.37U/mL。研究发现当固体发酵培养基中添加0.12%的吐温20(Tween 20)对菌株YN-2的CMCase活力影响最显著,在0.10% Tween 20的固体发酵培养基中菌株YN-2的FPA活力有所提高,在其他Tween 20添加浓度的固体发酵培养基中菌株YN-2的CMCase活力和FPA活力均受到不同程度的抑制。

Abstract:Strain YN-2 with high cellulase producing activity was obtained from soil of Yunnan farmland,and identified to be Penicillium oxalicum by analyzing its morphology and 18s RNA gene sequence.Experiments of measuring production and analyzing characters of cellulase were carried out after solid fermentation with Strain YN-2.The results showed that the highest activities of the carboxymethyl cellulose enzyme (CMCase 52.60U/mL) and the filter paper enzyme (FPAse 18.37U/mL) were obtained after the strain was cultivated with flask shake for 4d. The CMCase and FPAse activities were optimal at 50℃ with pH 4.8. The CMCase activity was enhanced in the presence of 0.12% Tween 20, and the FPAse activity in the presence of 0.10% Tween 20. However, with other concentrations of Tween 20, both CMCase activity and FPAse activity were inhibited.
