农杆菌介导转化甘蓝型油菜大田植株小孢子*

摘    要: 以甘蓝型油菜花油3号、湘油15号大田植株的小孢子为受体,进行了农杆菌介导转化,PCR检测证明获得了转基因植株。侵染比例为1.0,共培养时间为24h,植株生长阶段选择(植株再生后),农杆菌介导转化甘蓝型油菜小孢子的频率最高,花油3号为1.21株转基因植株/花蕾,湘油15号为0.51株转基因植株/花蕾。

Abstract:Using microspores from plants of Brassica napus cv. Huayou 3 and Xiangyou 15 on field as receptor, transformation mediated by Agrobacterium tumefaciens was carried out. The results of PCR indicates that transgenic plants have been obtained. When the proportion of A. tumefaciens and microspore was 1.0, and co-culture time was 24 h, and the selection was done after plantlets had been regenerated from embryoids, transgenic plants were obtained at highest frequency of 11 21 plants per flower bud for B. napus cv. Huayou 3 and 0. 51 plants per flower bud for B. napus cv. Xiangyou 15.
