摘    要:以贵州10种玉米基因型的幼胚为外植体,研究2,4-D浓度和基因型对玉米胚性愈伤组织诱导的影响,并进行再生体系研究。结果表明:不同基因型玉米幼胚诱导胚性愈伤组织的最适2,4-D浓度不同:黔单19、黔单20、黔单23、09H313、大青233、益玉6号母本、益玉6号父本的最适2,4-D浓度为1.0 mg/L,遵玉母本为2.0mg/L,黔单21为3.0 mg/L。另外,不同基因型胚性愈伤组织的诱导能力及分化再生能力差异明显。其中,胚性愈伤组织诱导率较高的是遵玉母本和大青233,分别为81.87%和71.39%,而绿芽分化率较高的是黔单23和黔单21,分别为89.80%和80.00%。贵州10种玉米基因型中黔单23、黔单21和益玉6号父本表现出较好的再生频率,可为农杆菌介导遗传转化提供备选受体。

Abstract:Immature embryos of ten maize (Zea mays L. ) genotypes from Guizhou Province were used to in- duce embryogenic callus. Effects of genotypes and 2,4-D concentrations on the induction of embryogenic cal- lus, as well as the regeneration ability were investigated in present work. The results showed that the optimal 2,4-D concentration for embryogenic callus induction were different depending on the genotypes. Addition of 1.0 mg/L 2,4-D gave the best effect in Qiandanl9, Qiandan20, Qiandan23, 09H313, Daqing233, the par- ent of Yiyu 6 and the male parent of Yiyu6, however the optimal concentrations for the parent of Zunyu and Qiandan21 were 2.0 mg/L and 3.0 mg/L, respectively. Also, induction and differentiation of embryogenic callus diversified among the genotypes. The best induction rate of embryogenic callus were investigated from the parent of Zunyu (81.87%) and Daqing233 (71.39%), and the highest rate of differentiation were ob- tained from Qiandan23 (89.80%) and Qiandan21 (80.00%). Among the ten genotypes, Qiandan23, Qi- andan21 and the male parent of Yiyu6 demonstrated the higher regeneration ability, thus may facilitate trans- formation via agrobacterium.
