磷酸盐还原为磷化氢功能菌株的筛选与鉴定

摘    要:利用厌氧培养反应瓶和筛选培养基以A2/O厌氧池、污泥浓缩池的污泥、养殖场新鲜猪粪、鸡粪、牛粪及鸭粪为种泥,通过跟踪厌氧培养过程培养液中总磷的去除率和吸收液中磷化氢的生成量筛选出最佳种泥,并对最佳种泥进行厌氧条件下能将磷转化为磷化氢的菌株进行了筛选、分离和鉴定.结果表明,6种泥中鸡粪的厌氧除磷能力最强,鸭粪能力最弱,污泥浓缩池中的污泥和猪粪次之,牛粪及厌氧池污泥有一定的作用.本实验条件下鸡粪是厌氧除磷工艺的最佳种泥,将成为今后生物除磷工艺厌氧除磷菌种增殖、驯化的首选污泥.在1周期7 d内鸡粪的最佳培养时间为5 d.鸡粪培养液经21 d培养后分离到2株肠杆菌科、2株真杆菌属和1株丁酸弧菌属,均是厌氧条件下磷酸盐还原为磷化氢的菌株,真杆菌属和丁酸弧菌属为首次分离到.

Abstract:Anaerobic reactor and screening medium were used to screen and isolate the strains, which could transform phosphorus as phosphine. The anaerobic sludge, the concentrated pool sludge, pig manure, chicken manure, cow manure and duck manure were prepared as inoculum. The suitable inoculum was estimated by determining the TP removal rate and the concentration of phosphine in the medium under anaerobic condition. The results indicated that chicken manure had the strongest activity of removing phosphate, followed by the concentrated pool sludge and pig manure, cow manure and anaerobic sludge also possessed certain activity, but duck manure had the weakest activity, so the chicken manure was the best inoculum and would be the first selection for reproduction and domestication of anaerobic phosphate-removal bacteria during the biological phosphate removal process. The optimal culture time for chicken manure was 5 days within a 7 days period. Two strains of Enterobacteriaceae spp., two of Eubacterium spp. and one of Butyrivibrio sp. were isolated from the chicken manure, and Enterobacteriaceae sp. and Eubacterium sp. were isolated anaerobic phosphate-removal bacteria for the first time.
