鸭胚骨骼肌成肌细胞中MyoD1和Myf5基因的表达与分析

摘    要:以高邮鸭和金定鸭为研究对象,采用实时荧光定量PCR方法测定生肌调节因子MyoD1和Myf5基因在不同发育阶段鸭胚(13、15、17、19、21和23胚龄)骨骼肌成肌细胞中的表达水平,分析MyoD1和尬庐基因在鸭胚胎期骨骼肌成肌细胞中的表达模式和差异.结果表明,Myf5和MyoD1基因在胸腿肌成肌细胞中呈现不同的表达规律:胸肌成肌细胞中,Myf5基因的表达呈“低→高→低→较高”的趋势,类似反“-”形;MyoD1基因的表达呈“低-高-低”的趋势,在金定鸭17胚龄时达到高峰后迅速下降(P〈0.01),并维持在低水平,类似“∧”形,而在高邮鸭17胚龄时达到高峰后维持高水平至19胚龄(P〉0.05),再下降并维持在低水平(P〈0.05),类似“∩”形.腿肌成肌细胞中,岣筘基因的表达呈“低-高-低”的趋势,类似“∧”形;MyoD1基因的表达呈“较高→低→高→低”的趋势,类似“-”形.结果提示:Myf5和MyoD1基因在骨骼肌成肌细胞中的表达具有时间和品种依赖性;两基因在两品种鸭胸腿肌中均在17胚龄时达到高峰,表明17胚龄可能是成肌细胞分化和增殖的一个重要时期.

Abstract:mRNA expression of myogenic regulatory factors MyoD1 and Mff5 gene in skeletal muscle cells of duck embryo at different stages (131 15, 17, 19, 21, 23 embryo age) was detected using real-time PCR, the expression patterns and differences ofMyoD1 and Mff5 gene in skeletal muscle cells of duck embryo were analyzed. The results showed that the Myf5 and MyoD1 gene ex-pression patterns in myobhtst of leg and chest muscle were different. The trend of Myf5 gene was similar to the anti-" -" shape inpectoral myoblasts of two duck breeds. In pectoral myoblasts, MyoD1 gene was similar to the " ∩" shape in Jinding duck, and wassimilar to the " ∧" shape in Gaoyou duck. In leg muscle myoblasts of two duck breeds, the expression trend of M3f5 gene was similax to the " ∧" shape , but MyoD1 gene was similar to the "-" shape. The breeds expression level of the two genes all reached thepeak at 17 embryo age, but MyoD1 gene of Gaoyou duck was maintained higher expression level in 19 embryo age, the others all declined sharply in 19 embryo age. The results suggested that the expression level of Myf5 and MyoD1 gene in myoblast was associatedwith development stage and varieties, and 17 embryo age was an important period of myoblast differentiation and proliferation.
