蝴蝶兰花梗组织培养技术研究

摘    要:对蝴蝶兰组织培养快繁技术的优化进行了较系统的研究,建立了蝴蝶兰花梗组织培养技术体系。主要包括:用腋芽启动的幼嫩花梗,以MS+3.0mg/L6-BA培养出芽率最高,达90%以上;MS+5.0mg/L6-BA+0.5mg/LNAA诱导茎尖原球茎状体,诱导率最高,达100%;原球茎状体增殖的最佳培养基为MS+3.0mg/L6-BA+0.2mg/LNAA+1.5%o活性炭,增殖系数达2。3;添加6-BA和NAA的1/2MS培养基有利于生根,平均根数达到3.2条;过渡培养能够提高蝴蝶兰组培苗移栽成活率。

Regulation for Technical Operations in Vitro Rapid Multiplication in Butterfly Orchids

Abstract:The improvement of in vitro rapid multiplication in butterfly orchids was systematically studied. Regulation for technical operations in vitro rapid multiplication in butterfly orchids was established. The main results were as follows. The experiments using the pedicle axillary bud, as explant showed that the highest efficiency of shoot formation was more than 90% in MS medium with 3mg/L 6 - BA;Shoot tips produced the higher percentage of PLB which the highest percentage up to 100% on MS medium supplemented with 5.0 mg/L 6 - BA and 0. 5mg/L NAA;The optimal medium for the proliferation of PLB was MS medium with 3mg/L 6 - BA , 0. 2 mg/L NAA and 1.5%0 activated carbon and the highest proliferation rate reached 2. 3; The 1/2MS medium with 6 - BA and NAA was beneficial for the rooting with a average rooting rate of 3. 2 ;The survival rate of plantlets was increased by transition culture in comparison with the control.
