都儿菜组织培养及快速繁殖的研究

摘    要:本试验以都儿菜的嫩茎为材料,对愈伤组织的诱导、不定芽的分化,不定芽的分化继代培养、试管苗的生根、试管苗移栽所需要的条件进行了研究,建立起都儿菜嫩茎的愈伤组织培养及快速繁殖技术体系.结果表明:MS+BA0.5mg.L-1+2,4-D1.4mg.L-1为诱导都儿菜嫩茎愈伤组织的理想培养基;MS+BA0.5mg.L-1是都儿菜愈伤组织和不定芽分化的理想培养基;1/2MS+IAA0.5mg.L-1是都儿菜不定苗生根的理想培养基;与实生苗相比,定植于田间的试管苗生长旺盛,肉质茎单位面积产量提高14%,单株增产24%.

Abstract:Using the Brassica oleracea tender stems as explant,tissue culture and rapid propagation of Brassica oleracea were established in order to study the different conditions of callus induction,bud differentiation,rooting,and transplantation of seedings.The results showed that the medium MS BA0.5mg.L-1 2,4-D1.4mg.L-1 was the best for inducing callus from stem,medium MS BA0.5mg L-1 was the most suitable for bud differentiation,medium 1/2MS IAA0.5mg L-1 was the optimum for rooting.The transplanted plantlets in the field grew vigorously by nursling plantlets effectively indoors,and the gain yield per unit area of fleshy stem could be up to 14% and the increment of 24% per plantlets was achieved.
