摘    要:为了探索最佳的染色体制片方法,以芝麻活体种子根尖为材料,研究根尖预处理方法、解离时间、取材长度和染色方法等关键因子对芝麻染色体制片的影响.结果表明,预处理方法为40 mg/L的放线茵酮溶液处理3h、饱和对二氯苯溶液处理4h或0.002 mol/L 8-羟基喹啉溶液处理3h;取材长度为0.5~1.0 mm;在V无水酒精∶...

Abstract:The germinating roots of sesame were used to study the effects of the pretreatment methods,dissociation time,sampling length of roots tips and staining on the chromosome preparation.The results showed that the optimal pretreatment methods were as follows:40mg/L cycloheximide for 3h,or saturated p-dichlorobenzene for 4h,0.002mol/L 8-Hydroxyquinolin for 3h;then dissociated with the solution(anhydrous alcohol: 37% concentrated hydrochloric acid=1∶1) for 10 min at room temperature;the sampling length of root tips was 0.5-1.0mm,and stained with modified carbolic acid-magenta solution for 8min.With this method a very good effect of chromosome preparation could be obtained.
