| 摘 要： | 以不同倍性染色体小麦种质为材料,探讨通过生长点分生组织进行染色体制片,获得理想染色体图形的方法.结果表明,自然条件下,上午9:00-10:00,切取幼苗基部分蘖节约2 cm,剥离茎外部组织,立即放入有冰水混合物的试管中,试管在0～1℃冰水中处理24～30h,之后将茎尖组织转入装有卡诺氏固定液(95％酒精∶冰醋酸=3∶1...

| Abstract: | The observation of chromosomes in plant cells is a foundation to study cytology.This study explored the method to get a clear image of chromosomes using stem tip meristems of different wheat germplasms with various chromosomal ploidy.The stem tip meristems about 2cm long at the seedling stage were pretreated in ice water mixture at 9:00-10:00 for 24-30h at 0-1℃,and then they were switched into ethanol-acetic acid fixative(3∶1) for three days.Finally,the materials were conserved in 70% ethanol.In this method to prepare samples,chromosomes with good dispersion and clear morphological characters could be observed.However,the success rate to obtain clear chromosome images was generally lower,compared with chromosome preparation method using root tip tissues.Therefore,the chromosome preparation method with stem tips could a useful complement to the method with root tips. |
