烤烟叶片叶绿素提取及其稳定性研究

摘    要:为了探讨不同提取剂对烤烟叶片叶绿素提取效率的影响以及维持烤烟叶绿素稳定性的最佳条件,以烤烟叶片叶绿素提取量为衡量指标,研究了7种提取剂及其不同用量对烤烟叶片叶绿素提取效果的影响,并测定了叶绿素在不同光照时间、不同温度下的稳定性。结果表明:烤烟叶片叶绿素提取效果最好的方法是80%丙酮和95%乙醇混合液(V乙醇∶V丙酮=1∶1)浸提法,最佳提取时间约为18h,最适的提取剂使用量为20mL,最佳提取温度为60℃。

Abstract:To compare the extraction efficiency of different extracting liquids of chlorophyll in flue-cured tobacco and explore the best conditions of keeping chlorophyll stable,this experiment studied the influence of seven different extracting methods on chlorophyll concentration,and tes-ted the stability of chlorophyll under different illumination time,temperature and solvent volume,taking the concentration of chlorophyll extract as index.The results showed that the extracting liquid mixed 1 volume of acetone with 1volume of ethanol was the best solvent for extracting chlorophyll.And the optimum extraction conditions were as follows: the chlorophyll was extracted in 20 mL extraction reagent for 18 hours at 60 ℃.
