摘    要:以大花卷丹为试材,以MS培养基为基础培养基,附加不同种类和质量浓度的植物生长调节剂(6-BA、NAA),研究大花卷丹离体培养快速繁殖技术。结果表明,鳞片组培快繁的最佳取材部位是中层鳞片。大花卷丹不定芽分化最佳培养基为MS+NAA 0.3mg/L+6-BA 1.5mg/L,其分化率可达76.7%。最佳增殖培养基为MS+NAA 0.2mg/L+6-BA 2.0mg/L,增殖系数达到4.91;最佳生根培养基为MS+IAA 0.2mg/L,生根率达到96.7%,平均每苗根数可达8.8条。移栽前期最佳的基质配比为河沙∶草炭土=1∶1,成活率达到76.0%。

Abstract:Using MS medium as basic medium,the influences of different concentration hormones(6-BA,NAA) on the tissue culture of Lilium leichtlinii were studied in order to explore the sui-table level of hormones and increase the efficiency of tissue culture.Transplantation of Lilium leichtlinii was also studied to establish a high-frequency propagation system.The result showed that the optimal explant was the scale in the middle and MS medium with NAA 0.3 mg/L and6-BA 1.5 mg/L was optimal for induction of adventitious shoots with an induction rate of 76.7%.The optimum multiplication medium was MS+NAA 0.2 mg/L+6-BA 2.0 mg/L,with the coefficient of multiplication being 4.91.The optimum rooting medium was MS supplemented with IAA 0.2 mg/L,with rooting rate of 96.7% and average root number of 8.8.Transplantation survival rate was high on the transplanting medium of sand∶pearlite=1∶1,up to 76.0%.
