摘    要:应用农杆菌介导的T - DNA转化技术,成功转化了生防绿木霉(Trichoderma viride)ZBS6,共获得368个转化子,转化效率为每106个分生孢子产生92个转化子.PCR和Southern blot分子鉴定以及遗传稳定性测定结果表明:转化子T - DNA插入率为90%,均能够稳定遗传,且T-DNA单拷贝插...

Abstract:In this study,Trichoderma viride ZBS6 was transformed based on agrobacterium-mediated T-DNA transformation technology and 368 transformants were obtained.The transformation efficiency was 92 transformants per 106 spores.The transformants were identified using PCR and Southern blot as well as genetic stability assay.The results showed that the percentage of positive transformants which inherited stably achieved as high as 90% and among them 80% were single-copy transformants.Further indoor biocontrol assay was conducted with Gaeumannomyces gramims var.tritici as test pathogen.Two transformants,T79 and T124,which showed obviously increased antagonism,were screened.This study laid the foundation for functional genomics study of T.viride and also supplied enough materials for insights into the molecular mechanism of antagonism.
