金顶侧耳胞外多糖测定方法的选择及提取工艺的优化

摘    要:比较了两种测定金顶侧耳胞外多糖的方法,即苯酚-硫酸法和蒽酮-硫酸法,结果表明,苯酚-硫酸法是测定金顶侧耳胞外多糖得量较为理想的办法,不仅稳定性好、精密度高,而且回收率也较理想。在此基础上,采用正交试验设计方法,并结合工业生产实际进行分析和验证,结果表明,提取金顶侧耳胞外多糖较为合适的主要工艺条件为:母液浓缩到原体积的1/4倍,用75%的乙醇醇析,醇沉10 h。

Abstract:This experiment was conducted to make a comparison between two following methods for determining Pleurotus citrinopileatus extracellular polysaccharides: the phenol-sulfuric acid method and anthrone-sulfuric acid method. The results showed that the phenol-sulfuric acid method was a better method for determining the content of extracellular polysaccharides, it not only had good stability and high precision, but also had high recycle rate. On this base, analysis and verification were performed by means of orthogonal design and production practice. The result indicated that the better appropriate extracting technology for Pleurotus citrinopileatus extraccellular polysaccharides was as follows: the fermental liquid was contrasted by 1/4, separating the extraccellular polysaccharides with 75% alcohol for 15 h.
