盐泽地分离的耐盐性芽孢杆菌菌株的分子鉴定

摘    要:采用生理生化检测、Rep-PCR指纹图谱分析、16S rDNA及gyrB基因序列分析技术,对在青海尕海盐泽地植物根际土壤中分离得到的几株菌落形态较为特殊的芽孢杆菌菌株进行鉴定。生理生化分析结果表明:分离的芽孢杆菌菌株革兰氏染色为阳性。Rep-PCR指纹图谱表明,不同分离菌株间的指纹图谱完全相同,为同种芽孢杆菌。16S rDNA及gyrB基因序列鉴定结果显示,菌株与地衣芽孢杆菌(Bacillus licheniformis)的亲缘关系最近。综合几种鉴定结果,最终将分离自盐泽地的芽孢杆菌菌株鉴定为B.licheniformis。

Abstract:Bacillus strains isolated from rhizospheres soil of plant in Gahai salt-land region of Qinghai province,which presented special branching-shape colony,were identified by physiological and bio-chemistry inspections,Rep-PCR fingerprints,16S rDNA and gyrB partial sequence analysis.The results showed that Bacillus strains were gram-positive.Rep-PCR fingerprints analysis suggested that strains were identified as one Bacillus spp..16S rDNA and gyrB gene sequence analysis results indicated that Bacillus strains presented closest affiliation with Bacillus licheniformis.Taken together,the Bacillus strains isolated from rhizosphere soil of plant in Gahai salt-land region were identified as B.licheniformis.
