黄瓜品种RAPD指纹图谱的构建及遗传相似性分析

摘    要:利用RAPD技术对22个具有广泛遗传基础的黄瓜品种进行了指纹图谱构建及遗传相似性分析,从240条RAPD随机引物中筛选出27条具有稳定多态性的引物.27条引物共扩增出163条带谱,其中多态性带70条,平均多态性比例为43.10%.9条RAPD引物产生的12个特异性标记可以单一地鉴别9个品种,利用不同引物的组合可将其他品种鉴别出来.通过UPGMA法进行聚类分析,当GSC=0.688 8时,可将22个黄瓜品种分为4个群.22个品种间的平均遗传相似系数为 0.800 4,说明黄瓜的遗传变异较低,遗传基础狭窄.

Abstract:Twenty-seven RAPD primers that could amplify stable polymorphic products were selected from 240 random primers for the fingerprinting construction of cucumber varieties and genetic similarity analysis.Totally 163 bands were amplified,among which 70 bands were polymorphic,the average percentage of polymorphic bands being 43.10%.Nine varieties could be identified independently with 12 specific markers generated by 9 primers.All the 22 varieties could be identified by the combination of several primers.The clu...
