玉米PEPC基因和抗白叶枯病基因Xa25的水稻聚合育种

摘    要:利用转玉米PEPC基因水稻HPTER(高感水稻白叶枯病)和抗白叶枯病水稻HX-3(具有抗白叶枯病基因Xa25)杂交,获得F1杂种,并对F1杂种进行花药培养,获得42株二倍体植株。通过对这些植株后代进行抗白叶枯病性鉴定,PEPC酶活性和光合速率测定及PCR检测,最后获得同时具有玉米PEPC基因和抗白叶枯病基因Xa25的水稻聚合株系7个,这些株系的农艺性状较HPTER有较大改善,可作为培育高产、抗白叶枯病水稻新品种的种质资源。

Abstract:A transgenic rice cultivar HPTER,which expresses a maize phosphoenolpyruvate carboxylase(PEPC)gene and is highly susceptible to bacteria blight disease(BB),was crossed with a BB resistant cultivar HX-3 containing a resistance gene Xa25.By anther culture from the F_1 hybrids of the cross,a total of 42 doubled-haploid(DH) regenerated plants were obtained.BB resistance evaluation,PCR analysis and detection of PEPC activity and photosynthetic rate in the DH line led to the identification of 7 pyramid DH lines containing both PEPC gene and Xa25 gene.The pyramid DH lines,with their agronomic traits greatly improved,may act as valuable germplasms for the development of new disease-resistant high-yielding rice varieties.
