秋水仙碱离体诱导薄皮甜瓜四倍体的研究初报

摘    要:以甜瓜组培苗茎尖为材料,研究了秋水仙碱诱导染色体倍性变异的最佳浓度、最适处理时间以及离体茎尖的最佳苗龄。结果表明:以8d苗龄的茎尖,在含有0.1mg/L秋水仙碱和较低浓度细胞分裂素的液体培养基中处理24-48h的诱导方法最有利于四倍体的产生,诱导染色体加倍的频率达30%~40%。变异株在田间表现出生长缓慢、叶片变宽加厚、茎增粗、节间变短、果皮变厚、种子变大增重的现象,显微镜观察变异后代根尖的染色体数目为2n=4x=48条。

Abstract:The experiment was carried out to induce and identify tetraploid of oriental melon by shoot tips in vitro. The effects of severial factors such as induced concentration, induced time and seedling age on the induction rates were studied. Results showed that the highest induction rate ranged from 30% - 40% under the optimized colchicines treatment combination of 0.1 mg/L colchicine for 24 h. In addition, the induced plantlet showed slower growth, smaller leaf index, stronger stem, shorter internode, thicker pericarp and bigger seed during the course of induction. The chromosome numbers of root - tip cells of the induced plantlets were 2 n = 4 x = 48.
