好氧反硝化细菌LKX-1菌株培养基与发酵条件的优化

摘    要:采用单因素试验法研究了蒙氏假单胞菌LKX-1菌株的发酵培养基组成和发酵条件,并采用正交试验法对发酵条件进行了优化。结果表明:适宜的发酵培养基配方为:麦芽糖2.0%,酵母粉2.0%,KH_2PO_4 0.15%,MgSO_4·7H_2O 0.10%;最适发酵条件为:发酵温度30℃,接种量1.0%,初始pH值7.0,装液量100 m L/瓶(三角瓶容量250 m L),发酵时间24 h;在此条件下,发酵液中活菌数可达4.5×10~(14)CFU/m L。

Abstract:The composition of fermentation medium and fermentation conditions were studied by using single-factor test, and the fermentation conditions were optimized by using orthogonal experiment. The results showed that the optimal fermentation medium were as follows: maltose 2%, yeast powder 2%, KH2PO40.15%, MgSO4·7H2O 0.10%, and the optimal fermentation conditions were as follows:fermentation temperature 30℃ , inoculation 1%, the initial pH 7.0, the liquid volume 100 mL/ bottle, fermentation time 24 h. Under these conditions, the viable count reached 4.5×1014 CFU/mL.
